Background
Monkeypox is a zoonotic disease caused by monkeypox virus (MPXV), which is a member of orthopoxvirus genus. A29L is an intracellular mature virus (IMV) surface envelope protein that is homologous to vaccinia virus (VACV) A27 and is widely conserved in the Poxviridae family. The A27L protein (p14) is a multifunctional protein that has been implicated in virus attachment, virus–cell fusion, cell–cell fusion, plaque size, and the formation of enveloped virions. The involvement of A27 in mature virus (IMV) inhibits the assembly of fusion protein A26. After MVs are endocytosed by vesicles, the acidic endocytic environment activates the dissociation of A26 protein from MVs, resulting in fusion of the viral membrane with the vesicle membrane. A27 interacts with the integral membrane protein A17, and then forms a protein complex with A26 through disulfide bond linkage to anchor the A26 protein to mature virus (MV) particles.
