Description

HotStart Tth DNA Polymerase contains high-performance Tth DNA Polymerase bound to a proprietary antibody, and it is supplied in an inactive state and has no polymerase activity at ambient temperatures. This prevents extension of nonspecifically annealed primers and primer dimers formed at low temperatures during PCR setup and the initial PCR cycle. The antibody is denatured in the initial DNA denaturation step of the PCR reaction, then the Tth DNA Polymerase activity is restored. It is a thermostable DNA polymerase with intrinsic reverse transcriptase (RT) activity. The enzyme is a highly processive 5´→3´ DNA polymerase and lacks 3´→5´ exonuclease activity (proof reading). It possesses a very efficient intrinsic reverse transcriptase (RT) activity in the presence of manganese ions and unlike other RT enzymes.
HotStart Tth DNA Polymerase (Glycerol-Free) is a glycerol free formulation of our regular HotStart Tth DNA Polymerase and is well suited for automated high throughput testing, lyophilization or where accurate pipetting of small volumes are crucial. HotStart Tth DNA Polymerase (Glycerol-Free) is available in different sizes and concentrations.

Features

Notes

The reverse transcription temperature of HotStart Tth DNA Polymerase is 60℃~70℃, and the reverse transcription time can be 15~30min.
This system is more suitable for RT reaction with specific primers, and the primer Tm value should be 60℃ or higher.
The reaction system contains Mn²⁺, so the fidelity of this system is reduced and it is not suitable for cloning, sequencing and other experiments with high fidelity requirements.
Before use, mix gently by inverting to avoid foaming, and centrifuge gently. When preparing and dispensing the reaction solution, use new (uncontaminated) pipette tips, Microtubes, etc. to avoid contamination as much as possible.
