Description
  • Interleukin-4 (IL-4) is a cytokine secreted by type II T helper cells (Th2 cells). It guides the monocytes to differentiate to DCs by inhibiting the growth of macrophages. Monocytes will differentiate to macrophages in a culture system without IL-4. IL-4 also reduces the expression of CD14 molecules on the cell surface, while the reduction in CD14 expression level is an important marker of the differentiation of monocytes into DCs.
  • The biological effects of IL-4 include stimulating the proliferation of activated B cells and T cells, differentiation of CD4+ T cells into type II T helper cells, and stimulation of Th2 secretion of ,IL-4 IL-5, IL-6, IL-10, and IL-13. IL-4 also plays a critical role in regulating humoral immunity and adaptive immunity. It induces the switching of the B cell antibody class to IgE and upregulates MHC II production.

Usage
  • In-vitro cultivation of various types of cell.
  • During the culture of DCs, the combination of IL-4 and GM-CSF (Cat. No.: GMP-CC79) directionally differentiate monocytes into immature DCs. The DCs cultured this way have strong antigen uptake and processing abilities but weak antigen presentation abilities. Therefore, sequentially use with TNF-alpha (Cat. No.: GMP-C008) to promote DCs maturing.

Measured in a cell proliferation assay using TF‑1 human erythroleukemic cells. The specific activity of Recombinant human IL-4 is ≥ 1.0 × 107 IU/mg, which is calibrated against the human IL-4 WHO International Standard (NIBSC code: 88/656)
Recommendations
  • （1）Make sure the product properly stored and none expired before using.
  • （2）Please use water for injection for reconstitution, to a concentration > 100 μg/ml.
  • （3）Sterile condition without pyrogen is required for reconstitution, re-aliquoting, dilution.
  • （4）After reconstitution, aliquoted solution vials should be stored at -70°C and minimize freeze-thaw cycles preventing denaturation of protein.
